The ASTM E2276-10 standard, developed by the American Society for Testing and Materials (ASTM), describes a test method for determining the effectiveness of hygienic hand washes and hand rubs against temporary bacterial flora on hands. This test method is not intended for use with surgical hand rubs or pre-operative skin preparations.
The application of this test method requires compliance with regulations for the protection of human subjects. The testing method should be performed by persons trained in microbiology in facilities designed and equipped to work with infectious agents at biosafety level 2.
This in vivo test method was developed with the aim of detecting the ability of hygienic hand washes or hand rubs to eliminate selected bacterial strains from the experimentally contaminated skin of the hands of adult subjects. Since the two thumb pads and all eight finger pads can be used in any test, it allows an entry control to be included in the testing process, checking for viable bacteria remaining after the vaccine has dried, eliminating bacteria after treatment, and evaluating the bactericidal effectiveness of the tested product. No more than 100 µL of test bacterial suspension is required to complete this test. The results of tests with this test method form the basis for validation tests using a full hand test protocol conforming to the ASTM E1174 standard.
The test method described in the ASTM E2276-10 standard relates to testing with bacteria, but is easily adapted to work with protozoa and bacteriophages. Test methods defined in ASTM E2613 for working with fungi and ASTM E1838 for working with viruses of human origin should be followed.
