The EN 1499 standard, a European standard, describes requirements and defines a test method to determine whether a product for hygienic handwashing reduces the release of transient microbial flora on hands when used to wash volunteers' artificially contaminated hands.

The application of this test method on human volunteers requires compliance with legal regulations in certain European countries or regions.

This standard applies to surgical handrub or handwash products for use in areas and situations where disinfection is medically indicated. The most common environments are:

Test organisms are artificially contaminated with volunteers' hands while testing. The number of test organisms released from fingertips into sampling fluids is assessed before and after hygienic hand washing. The ratio of the two values ​​obtained represents a measure for the antimicrobial activity of the product under test. The test is repeated on 12 to 15 volunteers to ensure the necessary precision. To compensate for external effects, it is compared with the reduction achieved by a reference handwashing performed with the same volunteers on the same day and under comparable environmental conditions.

A suitable neutralizer is verified prior to testing. Neutralizer is used as sampling fluid to recover test organisms after hygienic hand washing to ensure neutralization or suppression of bactericidal or bacteriostatic activity in sampling fluids.
