DIN 38409 German Standard Methods for Inspection of Water, Waste Water and Sludge - Effects and Parameters Characterizing Substances (Group H)
In general, multiple test methods and laboratory techniques are used to detect and measure various contaminants in waters, wastewater and sludge. Laboratory controls take into account method performance and data quality, ensuring results are comparable. Depending on the contaminant levels in wastewater, methods are adapted to higher or lower detection limits as needed. For example, if the levels of certain contaminants in the wastewater are high enough, small volumes of wastewater can be tested without additional concentration processes. Test methods include sample processing steps, use of laboratory controls, and application of biosafety measures to ensure data can be interpreted for public health use.

Sample processing, sample preparation, sample concentration and different measurement methods are applied in the testing of water and wastewater. The methods chosen at each step are tailored for use with wastewater, a complex and variable mixture of chemical and biological, and the performance of wastewater sample handling procedures is evaluated using appropriate laboratory controls.
Proper storage and preparation of wastewater samples ensures accurate measurements. Samples are cooled to 4 degrees immediately after collection and processed within 24 hours to increase surveillance benefit. If it cannot be processed within 24 hours, a matrix recovery check is performed on the sample before cooling it at 4 degrees or freezing at minus 20 or minus 70 degrees.
Both liquid effluent and primary sludge samples must be thoroughly mixed (homogenized) before removing portions of effluent collected for the downstream process. The homogenization process may include additional procedures to break up samples effluent solids and separate virus particles.
