The Standard Plate Count test evaluates both total aerobic bacterial and total molds and yeasts. It can be accomplished by plate counts, membrane filtration, pour plates, or spread plate methods. This test is suitable for all non-sterile products and provides a standardized way to determine the density of heterotrophic bacteria or the microbial content of the sample.

Crude plate count results for standard agar plates should be within 25 to 250 colony forming units (CFU) or reported as an estimate. The recommended count range for Aspergillus niger and other filamentous colonies is between 8 and 80 CFU per plate. If no colonies are found on the plates, the result should be reported less than the sample dilution.
All test monitors show the environment and test control defined by the warning and action levels described in the procedure. Colonies found on negative test monitor plates are compared to test plates for contamination in the test system. All observations are noted with data (eg representative colonies). Positive test monitors show growth when appropriate.
Procedure : A sample of the specimen or appropriate dilution(s) is plated three times on soybean casein digestion agar for fungi. The specimens are spread evenly with a sterile, bent glass rod. After the plates are allowed to dry at room temperature, they are inverted and incubated.
