With the test method described in the ASTM E2011-21 standard, virucidal activity for both handwash and handrub formulations is determined by comparing the reduction in virus infectivity from hands treated with a test article to hands treated with a control article. The ASTM E2011-21 test method is performed on healthy individuals with undamaged skin on their hands and lower arms. These individuals avoid using antimicrobial products for at least one week before the test. Immediately before testing, subjects' hands are washed for one minute with a mild, non-antimicrobial soap and then dried. To reduce the possibility of cell culture assay contamination while test samples are plated for evaluation, the hands are then decontaminated with ethanol and optionally rinsed with sterile water to remove any ethanol residue. A suspension of the selected test virus is prepared and spread evenly over all surfaces of the subjects' hands, then left to dry. When evaluating handwash formulations, the subject's hands are moistened with water after viral inoculation. The hands are then treated with conditions that reflect real-world usage conditions. Then the subject's hands are rinsed and the sample is taken.
